Description

A small molecule research reagent from STEMCELL Technologies, BMS 493 (synonym: BMS 204493) is described in the product information as acting on the retinoic acid receptor pathway. It functions as an inverse agonist of the pan-retinoic acid receptors (RARs), which are ligand-dependent transcription factors that regulate gene networks controlling cell growth, differentiation, survival, and death. The compound has been shown to increase nuclear receptor co-repressor (NCoR) interaction with RARs.

The chemical identity is 4-[(E)-2-[5,5-dimethyl-8-(2-phenylethynyl)-6H-naphthalen-2-yl]ethenyl]benzoic acid (CAS 215030-90-3), with a molecular formula of C29H24O2 and a molecular weight of 404.5 g/mol. It is supplied as a crystalline solid with a reported purity of ≥98%. Reported solubilities are ~0.2 mg/mL in a 1:4 solution of DMF:PBS (pH 7.2), ~25 mg/mL in DMSO, and ~30 mg/mL in DMF.

Published applications include effects on bronchial tubule formation in mice, inhibition of activation of immature dendritic cells, and inhibition of differentiation of leukemic blasts. The product is reported stable at -20°C as supplied and should be protected from prolonged exposure to light; for long-term storage a desiccant is recommended, and it is stated to be stable as supplied for 12 months from the date of receipt. A stock solution in DMSO at ≤60 mM is suggested. The Safety Data Sheet indicates the chemical is considered hazardous under the 2012 OSHA Hazard Communication Standard and is intended for laboratory use.
