This single-use sealing accessory from QIAGEN closes empty ports on the Column Holder of the QIAvac HiSpeed LS vacuum manifold, the system built for parallel processing of up to 6 HiSpeed Mega or Giga plasmid purification columns in the HiSpeed Plasmid Mega/Giga EndoFree workflow. Each Blind Plug sits in any luer slot not occupied by a Connection Adapter and VacValve, holding the manifold airtight while buffers, lysate, and wash solution are drawn through the resin bed under regulated vacuum. Operating pressures span -100 mbar during equilibration up to -300 mbar during lysate loading, so an unsealed position would collapse the vacuum circuit and interrupt flow. Operators scale the run from a single column to a full six-column setup by swapping plugs for Connection Adapters as needed, confirming that every Column Holder port is either actively engaged in purification or mechanically sealed before the lid is closed with the Cramps. The polymer body withstands the guanidine hydrochloride and isopropanol buffers typical of plasmid binding and wash steps, as well as Buffer QBT equilibration volumes of 35 mL (Mega tips) or 75 mL (Giga tips). The plug is inserted by hand into the Column Holder lid before each protocol and removed and discarded afterward. Shipped as a service spare for the five plugs that accompany a new QIAvac HiSpeed LS (QIAGEN catalog reference 19505), this consumable is meant to be restocked after several high-yield nucleic acid preparations. Store at room temperature in the original packaging. Pair with VacValve Tool, Lock Bolt, and Snap-on Connector to keep the manifold sealed across every run.
