Description

A small molecule epigenetic modifier from STEMCELL Technologies that acts as a very specific inhibitor of the G9a (IC₅₀ = 1.9 µM) and G9a-like (GLP, IC₅₀ = 0.7 µM) histone methyltransferases. These methyltransferases target the lysine 9 position on histone 3 (H3K9). The compound inhibits G9a and GLP by occupying the histone binding site, preventing interaction with histones (Chang et al.; Kubicek et al.). It is supplied as the trihydrochloride hydrate form of the molecule.

Chemical name is 2-(hexahydro-4-methyl-1H-1,4-diazepin-1-yl)-6,7-dimethoxy-N-[1-(phenylmethyl)-4-piperidinyl]-4-quinazolinamine, trihydrochloride, hydrate, with molecular formula C₂₈H₃₈N₆O₂ · 3HCl [XH₂O] and molecular weight 600.0 g/mol (anhydrous basis); CAS Number 1808255-64-2. The product is a crystalline solid with a purity of ≥ 98%. Reported solubility is ≤ 16 mM in water and ≤ 8 mM in DMSO.

Storage: stable at -20°C as supplied; protect from prolonged exposure to light. For long-term storage, store with a desiccant. To prepare a 5 mM stock solution in water, resuspend 1 mg in 333 µL of water; prepare stock solution fresh before use. As a general guide, storage in DMSO at -20°C is recommended, aliquoted into working volumes to avoid repeated freeze-thaw cycles. Avoid final DMSO concentration above 0.1% due to potential cell toxicity. Published application: enhances reprogramming of mouse embryonic fibroblasts or fetal neural progenitor cells to induced pluripotent stem cells without using c-Myc and SOX2 (Shi et al. 2008a; Shi et al. 2008b). For research use only; not intended for human or animal diagnostic or therapeutic uses unless otherwise stated.
