Description

An epigenetic modifier from STEMCELL Technologies that acts as a highly specific inhibitor of the G9a histone methyltransferase (IC₅₀ = 1.9 µM) and the related G9a-like (GLP) methyltransferase (IC₅₀ = 0.7 µM). Both enzymes target the lysine 9 position on histone 3 (H3K9), and inhibition is achieved by the compound occupying the histone binding site, thereby preventing interaction with histones. The molecule is supplied as its trihydrochloride hydrate form.

The compound is presented as a crystalline solid with the chemical name 2-(hexahydro-4-methyl-1H-1,4-diazepin-1-yl)-6,7-dimethoxy-N-[1-(phenylmethyl)-4-piperidinyl]-4-quinazolinamine, trihydrochloride, hydrate (CAS Number 1808255-64-2; molecular formula C₂₈H₃₈N₆O₂ · 3HCl [XH₂O]; molecular weight 600.0 g/mol on an anhydrous basis) and a reported purity of ≥ 98%. Reported solubility is ≤ 16 mM in water and ≤ 8 mM in DMSO.

As supplied, the product is stable at -20°C and should be protected from prolonged exposure to light; for long-term storage it should be kept with a desiccant. In published reprogramming studies, the compound has been used to enhance the reprogramming of mouse embryonic fibroblasts or fetal neural progenitor cells to induced pluripotent stem cells without the requirement for c-Myc and SOX2.
