Description

A small molecule research compound from STEMCELL Technologies, this WNT pathway activator functions by inhibiting glycogen synthase kinase 3 (GSK3). It is an analog of the GSK3 inhibitor BIO and potently inhibits both GSK3α and GSK3β (IC50 = 10 nM), while less potently inhibiting the cyclin-dependent kinases Cdk5/p25, Cdk2/A, and Cdk1/B (IC50 = 2.4, 4.3, and 63 µM, respectively). By blocking β-catenin phosphorylation and degradation, the compound permits activation of transcription of WNT pathway-controlled genes.

Supplied as a crystalline solid, the compound has a molecular formula of C₁₈H₁₂BrN₃O₃, a molecular weight of 398.2 g/mol, and a reported purity of ≥ 95%. It is also known as 6-bromoindirubin-3'-acetoxime.

Published applications include use as a differentiation agent in stem cell research. The compound has also been reported to inhibit CD8+ effector T cell differentiation and to suppress viral gene expression, conferring protection against herpes simplex virus type 1 (HSV-1) infection in oral epithelial cells. The product is stable at -20°C as supplied and remains stable for 12 months from the date of receipt when stored with a desiccant and protected from prolonged light exposure. Stock solutions can be prepared in DMSO at concentrations up to 25 mM, but the compound exhibits low solubility in aqueous media, and final DMSO concentrations above 0.1% in cell culture should be avoided due to potential toxicity.
