Supplied by STEMCELL Technologies as a 1 mg aliquot of lyophilized powder, this small-molecule inhibitor targets glycogen synthase kinase-3 (GSK3) and is widely used to manipulate Wnt/beta-catenin signaling in stem cell research. Also known as 6-bromoindirubin-3-prime-oxime, the compound competes for the ATP-binding pocket of GSK3, blocking phosphorylation of beta-catenin and stabilizing its nuclear accumulation.

In mouse embryonic stem cell (mESC) culture, BIO is added at 0.5-5 microM to serum-free defined media to maintain pluripotency and suppress spontaneous differentiation. Researchers also deploy it transiently during directed differentiation protocols toward cardiac, neural, and hematopoietic lineages.

For research use only, the powder is typically reconstituted in DMSO to a 10 mM stock and stored at -20 degrees C, protected from light, with aliquots recommended to limit freeze-thaw cycles. Each vial delivers a single-use quantity intended for routine mESC maintenance and short signaling-pulse experiments.
