Description

STEMCELL Technologies supplies this small molecule as a potent and selective ATP-competitive inhibitor of mammalian target of rapamycin (mTOR). It is described as an mTOR pathway inhibitor that inhibits both the mTORC1 and mTORC2 complexes as well as FKBP-12. Reported biochemical characteristics include an IC₅₀ of 0.8 nM, approximately 1000-fold selectivity for mTOR over all PI3K isoforms, and no activity against a panel of 260 kinases at concentrations up to 10 µM.

The product is supplied as a crystalline solid at a stated purity of ≥ 98%, with a molecular formula of C₂₅H₃₁N₅O₄ and a molecular weight of 465.5 g/mol. The CAS number is 1009298-09-2, and an alternative code listed is CCG-168. The chemical name is 5-[2,4-bis[(3S)-3-methyl-4-morpholinyl]pyrido[2,3-d]pyrimidin-7-yl]-2-methoxy-benzenemethanol. The compound is stable at -20°C as supplied when protected from prolonged exposure to light, and remains stable for 12 months from the date of receipt. Reported solubility limits are ≤ 2 mM in DMSO and ≤ 1 mM in absolute ethanol. The compound has low solubility in aqueous media; for use as a cell culture supplement, stock solutions should be diluted into culture medium immediately before use, and the final DMSO concentration should be kept below 0.1% to avoid potential cell toxicity. As an example preparation, to obtain a 1 mM stock solution in DMSO, 1 mg is resuspended in 2.15 mL of DMSO.

Published cancer research applications include inhibition of proliferation in A549 and H838 small-cell lung cancer cell lines in vitro and tumour growth from human tumour xenografts in mice after oral administration; effects on tamoxifen-resistant (TamR) and estrogen deprivation-resistant (MCF7-X) breast cancer cell lines; induction of apoptosis in Hep-2 laryngeal cancer cells; synergistic induction of apoptosis with ABT-737 in rhabdomyosarcoma cells via suppression of MCL1; and inhibition of proliferation and glycolysis in HeLa cervical cancer cells. The product is supplied for research use only.
