ArciTect Hu CRISPR Optimization Kit, AZ

Designed for the optimization of genome editing in human cells, this kit from STEMCELL Technologies can also serve as a positive control for experiments using the ArciTect CRISPR-Cas9 genome editing system in human cells. It combines ArciTect Human B2M sgRNA, ArciTect Cas9 Nuclease, and a fluorophore-conjugated antibody against beta-2 microglobulin (B2M), available in APC, PE, and FITC conjugate formats.

The workflow involves complexing the Cas9 nuclease with the B2M sgRNA, delivering the ribonucleoprotein complex into human cells, and culturing for 48 - 96 hours before assessing editing efficiency by flow cytometry with the included antibody. Because B2M is a ubiquitously expressed cell-surface protein, loss of B2M expression provides a rapid, quantitative readout of editing success and also allows additional evaluation such as viability monitoring and assessment of cell type-specific marker expression. The kit has been validated for use with the ArciTect line of genome editing products and has been shown to deliver editing in hPSCs, T cells, and CD34+ hematopoietic stem and progenitor cells.

The included anti-B2M antibody is a mouse monoclonal (Clone 35, IgG1) reactive with human B2M, supplied as an aqueous solution containing 0.5% bovine serum albumin (BSA) and 0.03% ProClin 300 and free of unconjugated fluorophore, and is intended for use in flow cytometry.
