Description

A cell-permeable phenoxazine-derivative inhibitor of AKT kinase phosphorylation from STEMCELL Technologies, with a reported IC₅₀ of ~1-2 µM. The compound blocks translocation of AKT after insulin-like growth factor 1 (IGF-1) treatment and is supplied as the hydrochloride salt of the molecule.

The chemical name is 2-chloro-N,N-diethyl-10H-phenoxazine-10-butanamine, monohydrochloride (alternative name: 10-DEBC hydrochloride; CAS 925681-41-0). It has a molecular weight of 381.3 g/mol, a purity of ≥ 95%, and appears as a crystalline solid. Reported solubility limits are ≤ 13 mM in PBS (pH 7.2), ≤ 30 mM in DMSO, and ≤ 40 mM in absolute ethanol. The product is stable at -20°C as supplied and should be protected from prolonged exposure to light.

Published applications include cancer research, where the inhibitor suppresses growth and induces apoptosis in human rhabdomyosarcoma cell lines, inhibits proliferation of breast cancer cell lines alone or synergistically with chloroquine, and reduces replication of Myxoma virus in human tumor cell lines. In disease modeling, it has been shown to induce autophagy in neurons and to be neuroprotective in a primary neuronal Huntington's disease cellular model.
