Description

Provided by STEMCELL Technologies, this small molecule is a cell-permeable, reversible inhibitor of mammalian sirtuin 2 (SIRT2), a nicotinamide adenine dinucleotide (NAD)-dependent histone deacetylase involved in neurodegeneration, aging, cell cycle progression, and tumorigenesis. AGK2 is classified as an epigenetic modifier and exhibits an IC₅₀ of 3.5 µM against SIRT2, with minimal activity against SIRT1 or SIRT3 (IC₅₀ > 50 µM; Outeiro et al.).

The compound is supplied as a crystalline solid with a purity of ≥ 98% (CAS 304896-28-4; molecular formula C₂₃H₁₃Cl₂N₃O₂; molecular weight 434.3 g/mol). It is stable at -20°C for 12 months from the date of receipt when protected from prolonged light exposure; for long-term storage the manufacturer recommends a desiccant. AGK2 is soluble in DMSO at ≤ 2.3 mM. The chemical name is (E)-2-cyano-3-[5-(2,5-dichlorophenyl)furan-2-yl]-N-quinolin-5-ylprop-2-enamide. The compound has low solubility in aqueous media, and for cell culture use the stock should be diluted into medium immediately before use, keeping final DMSO below 0.1% to avoid cell toxicity.

Published applications include activation of the NLRP3 inflammasome in mouse bone marrow-derived macrophages (Youm et al.), reduction of ALDH1+ cancer stem cells in primary breast cancer populations (Zhao D et al.), suppression of SIRT2-induced autophagy in human cancer cell lines (Zhao Y et al.), and protection of dopaminergic neurons from α-synuclein-mediated toxicity in in vitro and in vivo models of Parkinson's disease (Outeiro et al.). The product is hazardous and is supplied with a Safety Data Sheet. It is intended for research use only and not for human or animal diagnostic or therapeutic applications.
