Description
A synthetic peptide aldehyde bearing the PARP cleavage site DEVD, supplied as the trifluoroacetate salt, inhibits caspase-3 and caspase-7 in a reversible manner by interaction of the aldehyde group with the active site cysteine of these caspases. Inhibition constants reported by STEMCELL Technologies are Ki = 0.23 nM for caspase-3 and Ki = 1.6 nM for caspase-7.
The product is a crystalline solid with a stated purity of ≥ 95% and a molecular weight of 502.5 g/mol (chemical formula C20H30N4O11 • XCF3COOH). It is stable for 12 months from date of receipt when stored at -20°C, protected from prolonged light exposure, and stored long-term with a desiccant. Reported solubility values are PBS (pH 7.2) ≤ 19 mM, DMSO ≤ 45 mM, and absolute ethanol ≤ 55 mM.
Caspase inhibitors of this kind are used to investigate caspase activation and apoptosis. A published application noted by the supplier is that it blocks camptothecin-induced apoptosis in bone tumor cells.
