Description

From QIAGEN, Uracil-N-Glycosylase (UNG) is an enzyme that removes deaminated cytosine bases from DNA. It is supplied as a kit component for the automated purification of genomic DNA from formalin-fixed, paraffin-embedded (FFPE) tissues, where it is incorporated into UNG-containing kits such as the EZ1&2 DNA FFPE UNG Kit and the QIAamp DNA FFPE Advanced UNG Kit.

Formalin fixation, paraffin embedding, and long-term storage of FFPE tissue samples can introduce the most common sequence artifact: deamination of cytosine bases to uracil. This leads to a C→T conversion during amplification and false results in sensitive downstream analyses such as Next-Generation Sequencing (NGS) or digital PCR (dPCR). Including UNG in the workflow removes these deaminated cytosine bases before DNA binding, helping to prevent such artifacts in DNA sequencing analyses.

UNG is shipped on dry ice and should be stored immediately upon receipt at –30 to –15°C in a constant-temperature freezer. Under these conditions, the enzyme is stable until the expiration date printed on the UNG tube label. The products incorporating UNG are intended for molecular biology applications and are not intended for the diagnosis, prevention, or treatment of a disease.
