Description

QIAGEN Proteinase K is a broad-spectrum, subtilisin-related serine protease derived from the fungus Parengyodontium album (Tritirachium album). The recombinant 28.9 kDa enzyme is expressed in Komagataella phaffii (Pichia pastoris) and is extensively purified for high solubility, specific activity and low residual DNA content. It is supplied as a ready-to-use solution at a concentration of 20 mg/mL with an activity of >600 mAU/mL, and is certified RNase-free and DNase-free.

The enzyme remains active across a wide range of temperatures, pH values and denaturing conditions, including in the presence of SDS and EDTA, and is particularly effective between 50–65°C across a pH range of 7.5–12.0. This high specific activity enables efficient digestion of proteins, including DNases and RNases, during nucleic acid preparation without compromising DNA or RNA integrity, making it well-suited to short digestion times. The ready-to-use solution is optimized for use in QIAGEN kits and is shipped at ambient temperature.

QIAGEN Proteinase K is stable for up to 1 year after delivery when stored at room temperature; storage at 2–8°C is recommended to prolong shelf-life. Typical applications include the isolation of genomic, plasmid and high molecular weight DNA and RNA, as well as the digestion of structural proteins and chromatin.
