Description

From QIAGEN, this proteinase K is supplied as a ready-to-use solution for proteolytic digestion during genomic DNA purification. The reagent facilitates the removal of proteins and nucleases, supporting improved DNA quality for downstream applications such as PCR and sequencing, and is a molecular biology grade protease intended for efficient protein degradation.

In QIAGEN's Puregene DNA workflows, the enzyme is listed as an optional user-supplied reagent for protocols involving body fluid and buccal cell samples. The Puregene kits purify high-molecular-weight genomic, mitochondrial, or viral DNA from various sample types by lysing cells with an anionic detergent in the presence of a DNA stabilizer that limits DNase activity. RNA is removed by an RNA-digesting enzyme, additional contaminants such as proteins are removed by salt precipitation, and genomic DNA is recovered by alcohol precipitation and dissolved in hydration solution (1 mM EDTA, 10 mM Tris·Cl pH 7.5). Purified DNA typically shows an A260/A280 ratio between 1.7 and 1.9 and can be up to 200 kb in size.

Users should follow the manufacturer's instructions in the product insert, including recommended temperatures and safety precautions.
