Description

A recombinant DNA polymerase from QIAGEN, originally isolated from Thermus aquaticus and expressed in E. coli, is supplied under a licensing agreement with Hoffmann-La Roche for standard and specialized PCR applications such as differential display and PCR-based DNA fingerprinting. The enzyme is provided with QIAGEN PCR Buffer, which is formulated to minimize the need for optimization of primer–template systems, and with CoralLoad PCR Buffer, a 10x concentrate containing gel-tracking dyes that allows PCR products to be loaded directly onto an agarose gel without adding loading buffer.

The enzyme has a concentration of 5 units/µl, an extension rate of 2–4 kb/min at 72°C, and a half-life of 10 minutes at 97°C or 60 minutes at 94°C. It possesses 5'→3' exonuclease activity and adds a terminal A overhang, while lacking 3'→5' exonuclease activity as well as contaminating nuclease, RNase, protease, and self-priming activities. Q-Solution, a 5x additive also included, helps amplify difficult templates such as GC-rich sequences by modifying the melting behavior of DNA. The buffers contain 15 mM MgCl₂, providing a final concentration of 1.5 mM in the standard reaction mix.

The product is shipped on dry ice and should be stored at –30 to –15°C in a constant-temperature freezer immediately upon receipt. It is intended for molecular biology applications and is not for use in the diagnosis, prevention, or treatment of disease. Comprehensive quality control testing is performed on every lot, including PCR specificity and reproducibility assays with low-copy targets amplified from human genomic DNA.
