Description

A recombinant endonuclease from QIAGEN, derived from psychrophilic bacteria and produced in Escherichia coli, this 39.4 kDa enzyme is highly effective at digesting diverse DNA and RNA substrates. It maintains robust activity under challenging conditions, including elevated salt concentrations, extremes of pH, and temperature variations, making it valuable for removing unwanted nucleic acid contaminants during protein purification in both laboratory and biomanufacturing workflows.

The enzyme is supplied at a concentration of ≥250 U/µL in 20 mM Tris (pH 7.1 at 25°C), 620 mM NaCl, 26 mM MgCl₂, and 50% (v/v) glycerol. One unit is defined as an increase in absorbance at 260 nm of 1.0 in 30 minutes at 37°C in 50 mM Tris-HCl buffer, pH 8.5 (at 25°C), supplemented with 500 mM NaCl, 5 mM MgCl₂, 0.1 mg/mL BSA, and 0.5 mg/mL herring sperm DNA as substrate. For optimal activity, a minimum of 1 mM Mg²⁺ ions is required.

For general guidance, a range of 5–100 units per 1 mL of reaction mixture or lysate is recommended, with incubation at 25–37°C for 30–60 minutes. The product is shipped on dry ice and should be stored at −20°C. Inactivation can be achieved by incubation at 45°C for 30 minutes or at 50°C for 15 minutes with reducing agents such as DTT (2–5 mM); at least 1 mM DTT is required for complete inactivation.
