Description

A recombinant endonuclease derived from psychrophilic bacteria and produced in E. coli, Saltonase from QIAGEN is engineered for nucleic acid removal in biopharmaceutical production. It is highly active across a wide range of salt concentrations and pH levels, effectively digesting both DNA and RNA substrates under varied buffer conditions and temperatures, making it well suited for eliminating undesired nucleic acid contamination during protein purification in laboratory and biomanufacturing workflows.

Saltonase is offered in two purity grades—Molecular Biology Grade (MBG) and GMP-grade—both supplied at a concentration of ≥250 U/μL. MBG material shows ≥95% purity by SDS-PAGE, while GMP-grade is validated to ≥99% purity by HPLC and SDS-PAGE. GMP-grade specifications include endotoxin levels ≤0.25 EU per 1000 U and bioburden ≤10 CFU per 100,000 U, with no protease activity detected. QIAGEN recommends a working concentration of 20–50 U/mL. The enzyme retains activity across 0–900 mM NaCl, with peak performance at 500 mM NaCl, pH 8.5 and 37°C, where high salt tolerance helps reduce aggregation and viscosity during cell lysis in viral vector production. It cleaves DNA into 3–5 nucleotide fragments.

The enzyme is supplied in 20 mM Tris (pH 7.1 at 25°C), 620 mM NaCl, 26 mM MgCl2 and 50% (v/v) glycerol. One unit is defined as an increase in absorbance at 260 nm of 1.0 in 30 minutes at 37°C in 50 mM Tris-HCl buffer, pH 8.5 (25°C) supplemented with 500 mM NaCl, 5 mM MgCl2, 0.1 mg/mL BSA and 0.5 mg/mL herring sperm DNA as a substrate. QIAGEN GMP-grade enzymes are manufactured in compliance with ISO 13485, US QSR (21 CFR 820) and ISO 9001, are free from animal-origin materials, and contain no beta-lactam antibiotics, supporting biomanufacturing workflows from early development through clinical and commercial production.
