Description

QIAGEN's real-time PCR thermal cycler combines a 384-well block with a 5plex fiber-optic detection system for quantitative PCR applications such as gene expression analysis, genotyping and pathogen detection.

The instrument pairs a fast thermal block with a sensitive fiber-optic shuttle. A full cycling run can be completed in as little as 30 minutes, and an entire 384-well plate is read in approximately 6 seconds regardless of the number of filters used. The aluminum special-alloy sample block accepts sample volumes from 2 to 30 µl (5 to 20 µl recommended) and is compatible with 384-well PCR plates sealed with optical film. A motorized heated lid is adjustable from 30°C to 110°C and applies automated 30 kg contact pressure.

Thermal performance includes a 3°C to 99°C range, ±0.1°C accuracy and uniformity of ±0.15°C at 55°C, ±0.25°C at 72°C and ±0.50°C at 95°C measured 15 seconds after the clock starts. Maximum heating rate is 4°C/s (average 3.8°C/s) and maximum cooling rate is 2°C/s (average 1.7°C/s), with a 25°C temperature gradient in 0.1°C increments. The system detects 1 nmol/l FAM at a 30 µl sample volume in a 384-well plate, with a 10-log-stage dynamic range. Excitation is provided by four high-intensity LEDs (blue, green, white, red) and emission by a photomultiplier (PMT), using color filter modules at 465±15 / 524±12 nm (blue: FAM, SYBR Green, EvaGreen), 510±15 / 565±15 nm (green: JOE, HEX, VIC), 560±15 / 610±15 nm (orange: ROX, Texas Red), 625±10 / 680±15 nm (red: Cy5, Alexa Fluor 647) and 625±10 / 710±20 nm (NIR 1: Cy5.5, Quasar 705). Analysis methods include absolute and relative quantification, ΔΔCt, allelic discrimination, efficiency calculation, DNA melting curves and endpoint POS/NEG analysis, with export to Excel, CSV, LIMS, GenEx and qBase. The cycler is PC-controlled under Windows 7/8/10, weighs 30 kg, measures 59 × 27.5 × 27.5 cm (70 × 27.5 × 50 cm when opened), runs at a 45 dB noise level, draws up to 850 W and operates on 230 V at 50–60 Hz.
