Used as a component of QIAGEN Plasmid Kits, this gravity-flow, anion-exchange tip from QIAGEN supports the purification of transfection-grade plasmid DNA. The prepacked tip operates by gravity flow and never runs dry, helping to minimize hands-on time during plasmid preparation. In the standard workflow, bacterial lysates are cleared by centrifugation before the cleared lysate is loaded onto the tip.

At the Maxi scale, the tip processes 100–500 ml of culture as starting material with manual, gravity-flow operation based on anion-exchange technology. Each run is designed for 1 sample per run, with a variable elution volume and a time per run of 160 min. The expected yield is <500 µg of high-copy plasmid DNA, and low-copy plasmids as well as cosmid DNA can also be purified.

Plasmid DNA obtained with QIAGEN tips is suited to applications such as transfection, cloning, sequencing, and capillary sequencing. The unique anion-exchange resin delivers DNA purity equivalent or superior to that obtained by two successive rounds of CsCl gradient centrifugation. The purification system is formulated to avoid toxic substances including phenol, chloroform, ethidium bromide, and CsCl, lowering hazard to both user and environment.
