Plastic gravity-flow anion-exchange tips from QIAGEN, supplied as a component of the QIAGEN Plasmid Kits range for purification of up to 10 mg of transfection-grade plasmid or cosmid DNA. The unique anion-exchange resin is developed exclusively for nucleic acid purification, delivering DNA purity equivalent or superior to that obtained by two successive rounds of CsCl gradient centrifugation.

Prepacked tips operate by gravity flow and never run dry, minimizing hands-on time, while the entire plasmid purification system avoids toxic substances such as phenol, chloroform, ethidium bromide, and CsCl. Bacterial lysates are cleared by centrifugation and loaded onto the tip, where plasmid DNA selectively binds under low-salt and pH conditions. RNA, proteins, metabolites, and other low-molecular-weight impurities are removed by a medium-salt wash, and pure plasmid DNA is eluted in high-salt buffer.

The DNA is concentrated and desalted by isopropanol precipitation and collected by centrifugation. Processing is manual by gravity flow, supporting high-copy, low-copy, and cosmid DNA. Purified plasmid DNA is highly suitable for transfection, cloning, sequencing, capillary sequencing, and in vitro transcription. Manufactured in Germany.
