This key reagent supports the targeted amplification step within the QIAseq NGS library preparation workflow from QIAGEN. PAP Enzyme performs the primer extension and molecular barcode integration that follows adapter ligation on fragmented DNA or RNA-derived templates. Packaged as a single-use vial, the enzyme is supplied in an optimized buffer formulation ready for direct incorporation into standard library construction protocols.
The enzyme is paired with QIAseq primer mixes and indexed adapters to generate sequencing-ready libraries for Illumina platforms. Researchers typically add 1-2 µL of PAP Enzyme per reaction during the targeted enrichment step, which captures regions of interest using molecular-barcoded primers. The resulting libraries are compatible with downstream cluster generation, sequencing, and bioinformatics analysis.
Storage recommendations require freezing at -20°C to preserve enzymatic activity across multiple freeze-thaw cycles. Each vial contains sufficient enzyme for approximately 96 reactions depending on library complexity and template input. For research use only and not intended for diagnostic procedures, this reagent supports reproducible library yields when combined with QIAseq targeted panels and accessory enzymes used in the same workflow.
