A T4 bacteriophage-derived ligase from QIAGEN, this enzyme catalyzes phosphodiester bond formation between adjacent 3'-hydroxyl and 5'-phosphate termini of double-stranded DNA, supporting both blunt-end and cohesive-end (sticky-end) ligation reactions in next-generation sequencing workflows. Supplied as a ready-to-use liquid formulation in a single vial, the ligase integrates into standard NGS library preparation protocols, including adapter ligation to fragmented genomic DNA, PCR-free library construction, and circularization steps for specialized capture or rolling-circle applications. The enzyme is qualified for use with QIAGEN NGS reagent systems and is compatible with fragmented gDNA, amplicons, and synthetic oligonucleotide substrates. Activity is preserved when reactions are assembled on ice and incubated at 16-25 °C for 10-30 minutes, depending on substrate geometry. The product arrives frozen and should be stored at -20 °C; avoid repeated freeze-thaw cycles by aliquoting upon first use. Typical reaction volumes of 20-50 µL accommodate standard library preparation scales from nanogram to microgram input. The ligase is intended for research use only and is suitable for in vitro molecular biology and sequencing applications.
