The QIAseq UPX kit streamlines ultra-low-input transcriptomics by converting RNA from a single cell or a few picograms into Illumina-ready libraries through a streamlined single-tube workflow. Built on random-primed reverse transcription followed by targeted amplification, the chemistry tolerates degraded or limited starting material that conventional poly-A capture struggles with, making it well suited for laser-capture microdissections, sorted rare populations, and fixed tissue specimens in research use only workflows.

Each kit supplies pre-formulated enzyme mixes, reaction buffers, and indexed adapters in single-use vials, supporting one sample through cell lysis, cDNA synthesis, pre-amplification, indexing, and final library cleanup. A typical input of 10 pg to 10 ng total RNA yields sequencing-ready cDNA with low drop-out rates, and the protocol recommends storing reagents at -15 to -25 degrees C to preserve enzymatic activity. Researchers performing single-cell RNA-seq, whole-transcriptome profiling of clinical biopsies, or low-abundance microbial transcriptomics rely on this QIAGEN workflow to deliver reproducible insert sizes and unbiased gene coverage on Illumina sequencing platforms.
