UltraRun LongRange PCR Kit (100)

QIAGEN provides a kit for ultrafast, hot-start mediated long-range PCR. The ready-to-use master mix combines hot-start controlled Taq polymerase, a proofreader and other additives, enabling amplification of up to 30 kb from any DNA or cDNA template. A combined 30 seconds/kb annealing-extension step delivers high yields from as little as a few picograms of template, supporting long-range PCR with subsequent analysis in a day. The antibody-mediated hot-start mechanism keeps both polymerase and proofreader activities inactive until heat activation at 93°C, preventing primer or DNA damage from premature enzymatic activity, while the uracil-literate proofreader exhibits 10x higher fidelity than Taq.

The 4x-concentrated master mix format allows greater flexibility for higher template input volumes and direct loading onto agarose gels. Moderate multiplexing of up to 6 targets is supported across approximately 1–9 kb. Optional orange and blue tracer dyes provide visual pipetting control: when blue-stained template is added to orange master mix, the colour shifts to green. These dyes run at approximately 50 bp (orange) and 4000 bp (blue) on a 1% agarose gel.

The kit includes Q-Solution, an optional non-toxic PCR additive used at one working concentration, which facilitates amplification of GC-rich and other challenging templates. Reactions can be fully set up at room temperature without ice and are compatible with automated pipetting systems such as the QIAgility instrument. The product is suited for long amplicon sequencing in immunology, cancer and microbial workflows, including analysis of oncogenes, HLA loci, and pan-bacterial and fungal sequences such as 16S, 23S and ITS.
