Description
A recombinant DNA polymerase from QIAGEN, derived from the thermophilic eubacterium Thermus aquaticus, delivers fast PCR setup with minimal optimization for efficient amplification of routine, low-copy, longer or difficult targets. The enzyme is supplied at a concentration of 5 units/µL and features 5'→3' exonuclease activity for removing RNA primers, while lacking 3'→5' exonuclease proofreading activity. Thermostable activity above 70°C makes it suitable for standard and specialized PCR amplification applications, with a temperature optimum of 72°C and no denaturation at 95°C.
Key specifications include an extension rate of 2–4 kb/min at 72°C, a half-life of 10 min at 97°C and 60 min at 94°C, and an amplification efficiency of ≥10^5 fold. A non-template-dependent terminal transferase activity adds a single unpaired adenosyl residue to the 3'-end of amplified products (extra A addition), supporting efficient TA cloning. Applications include PCR, RT-PCR, DNA fingerprinting, and TA cloning, using genomic DNA and cDNA targets in endpoint PCR amplification reactions.
The product is supplied with QIAGEN PCR Buffer formulated for fast setup with minimal optimization, CoralLoad PCR Buffer containing two gel-tracking dyes for immediate loading onto agarose gels, Q-Solution to facilitate amplification of GC-rich templates or those with a high degree of secondary structure, and a separate 25 mM MgCl2 solution for optimization. The standard PCR Buffer provides a final MgCl2 concentration of 1.5 mM. Performance features include lot-to-lot reproducibility with low-copy targets, specific amplification of PCR products longer than 5 kb, a wide annealing temperature window of 50–60°C without optimization, tolerance of variable magnesium ion concentrations from 1.5 to 4.0 mM, and tolerance of primer Tm value differences greater than 25°C without optimization. Products are shipped on dry ice and retain full activity at room temperature (15–25°C) for 2 weeks; they should be stored at –20°C in a constant-temperature freezer upon receipt.
