EpiTect PCR Control DNA Set (100)

This QIAGEN set provides validated control DNAs designed for methylation analysis workflows, serving as positive and negative references in end-point methylation-specific PCR (MSP) and real-time methylation-specific PCR applications. The controls help monitor bisulfite conversion efficiency and overall assay performance, and are suitable for assay validation, standard curves, and quality control.

The set includes methylated and unmethylated control DNAs (both pre-bisulfite converted). The standard concentration is 10 ng/µl (1 µg in total), except for the unmethylated control DNA, which is supplied at 50 ng/µl (10 µg in total). When used with primer sets targeting unmethylated unconverted genomic DNA, unmethylated bisulfite-converted target DNA, and methylated bisulfite-converted target DNA, the controls produce defined expected PCR results, allowing verification of each assay condition alongside no-template controls. Methylated and unmethylated control DNAs can be mixed to generate 100%, 90%, 50%, 10%, and 0% methylated standards for quantification of unknown samples, for example with EpiTect MethyLight Assays run on the human PITX2 gene.

The control DNAs can also be used as standards in real-time methylation-specific quantification. The DNA is derived from human sources, so primers for mouse or rat will not bind. Bisulfite-treated DNA prepared with EpiTect Bisulfite Kits has been shown to remain stable for three years at –20 °C before use in methylation-specific PCR.
