From QIAGEN, this ready-to-use mastermix is specifically developed and functionally validated for reliable mutation analysis through multiplex PCR. Designed for endpoint PCR amplification of genomic DNA, the kit is based on the highly specific HotStarTaq Plus DNA Polymerase paired with a proprietary buffer system that ensures robust multiplex amplification of mutation sites without the need for optimisation.

The mastermix contains HotStarTaq Plus DNA Polymerase together with MgCl2, dNTPs, and an innovative PCR buffer purpose-built for multiplex mutation detection and preamplification of genomic SNP loci. A unique additive called Factor MP, combined with balanced salts and additives, ensures comparable annealing and extension efficiency across all primers in the reaction, enabling reliable high-yield multiplex amplification of multiple fragments in parallel.

The Type-it Mutation Detect PCR Kit is suited to detecting deletions, insertions, and translocations, and is also well suited for multiplex PCR-based preamplification of SNPs ahead of genotyping systems such as Applied Biosystems' SNaPshot. The kit is supplied with Q-Solution, an innovative PCR additive that facilitates amplification of difficult templates, and CoralLoad Dye, a gel-loading reagent containing tracking dyes that allows amplification products to be loaded directly onto agarose gels or the QIAxcel Advanced System without additional loading buffer. Dedicated protocols are provided for various downstream detection platforms, including agarose gels, the QIAxcel System, Agilent BioAnalyzer, and capillary sequencers, along with recommendations on template amount, cycle numbers, and PCR conditions. Reactions can be assembled at room temperature for greater convenience and ease of use.
