Description

This QIAGEN kit is designed for fast and reliable multiplex PCR analysis of microsatellite loci. It is functionally validated for microsatellite analysis and provides reliable genotyping of humans, animals, plants, and bacteria using microsatellite, STR, or VNTR markers, without the need for lengthy optimization procedures.

The kit is based on proprietary QIAGEN multiplex technology and includes HotStarTaq Plus DNA Polymerase, a chemically modified hot-start enzyme, along with a specially developed multiplex buffer containing Factor MP. The buffer provides a balanced combination of KCl and (NH4)2SO4 salts and additives that ensure comparable efficiencies for annealing and extension of all primers in the reaction. Factor MP increases the local concentration of primers at the DNA template and stabilizes specifically bound primers, while stringent primer-annealing conditions are maintained over a wider range of temperatures and Mg2+ concentrations than conventional PCR buffers. The kit also includes Q-Solution, a PCR additive that modifies the melting behavior of DNA to facilitate amplification of difficult templates.

Optimized for endpoint multiplex PCR amplification of genomic DNA, the product enables reaction setup at room temperature and ensures high product yields for all amplicons up to 500 bp long. It is validated for use with fluorescent primers on high-resolution capillary sequencers, as well as agarose gels, the Agilent Bioanalyzer, and the QIAxcel Advanced System. Dedicated application-specific protocols are included for the amplification of microsatellites and STRs, along with guidance on template amounts, cycle numbers, and PCR conditions. The kit is intended for microsatellite, STR, SSR, and VNTR analysis across diverse research fields.
