Type-it Fast SNP Probe PCR Kit (4000)

A ready-to-use master mix system from QIAGEN designed for probe-based SNP genotyping using 5'-nuclease probe chemistry. The 2x master mix contains HotStarTaq Plus DNA Polymerase — a chemically modified hot-start enzyme activated by a 5-minute, 95°C incubation step that prevents formation of misprimed products and primer–dimers during reaction setup — along with a novel fast-cycling PCR buffer incorporating Q-Bond technology, a 5x Q-Solution additive, ROX passive reference dye, a dNTP mix, and RNase-free water.

The buffer is specifically formulated for sequence-specific 5'-nuclease probes and provides highly stringent binding of the allele-specific match probe, producing wide and clear separation of allele clusters for unambiguous SNP allelic discrimination. According to the manufacturer's handbook, dense clusters are obtained even with 1 ng of template DNA. Q-Solution, already included at a defined concentration in the 2x master mix and supplied separately for greater flexibility, modifies the melting behaviour of DNA to improve amplification of GC-rich or secondary-structure-prone templates and difficult SNP loci.

The kit is functionally verified with commercially available SNP genotyping assays and is compatible with TaqMan MGB, TaqMan, and other dual-labeled probes. It is shipped on dry ice and should be stored immediately upon receipt at –20°C; the master mix can alternatively be stored at 2–8°C for up to 2 months without reduction in performance. The product is intended for molecular biology applications and is not intended for the diagnosis, prevention, or treatment of a disease.
