The diazo dye penetrates only cells with compromised plasma membranes, leaving live cells colorless and refractile, which enables manual enumeration on a hemocytometer or automated counting on instruments such as the Countess and TC20. Researchers typically mix a 1:1 or 1:10 ratio of trypan blue with a cell suspension and load 10 uL onto the counting chamber, scoring viable versus non-viable cells within three to five minutes to prevent further dye uptake from skewing the result.

Common applications include passaging adherent cell lines, calculating seeding density for culture dishes, post-thaw viability checks after cryogenic storage, and quality control steps in cell therapy and stem cell workflows.
