CoV Protease Substrate-1 EDANS,1000tests

A fluorescence resonance energy transfer (FRET)-based peptide substrate from STEMCELL Technologies, designed for the detection of coronavirus (CoV) protease activity. The coronavirus main protease, also called 3CLpro, regulates the CoV replication complex and is a potential target for therapeutic applications. The substrate consists of a 14-amino-acid sequence (KTSAVLQSGFRKME) carrying the DABCYL quencher at the N-terminus and the EDANS donor at the C-terminus. When intact, favourable energetic overlap of the EDANS excited state with the DABCYL absorption produces efficient intramolecular fluorescence quenching; cleavage by CoV proteases separates the two fragments and restores EDANS fluorescence in direct proportion to protease activity.

Key specifications include a predicted molecular mass of 2584.98 kDa, an excitation wavelength of 336 nm, an emission wavelength of 455 nm, a cutoff of 420 nm, an extinction coefficient of 5900 cm⁻¹ M⁻¹, and a purity of ≥ 95%. The product is supplied as a lyophilised solid prepared from a dimethylformamide (DMF) solution by vacuum drying. According to the manufacturer's safety data sheet, the substance is not classified as hazardous under the 2012 OSHA Hazard Communication Standard (29 CFR 1910.1200); it is described as a red solid that is not regulated for transport under DOT, TDG, MEX, ICAO (air), IATA, IMDG, RID, ADR, or ADN.

For preparation, centrifuge the vial before opening and reconstitute in DMSO without vortexing; store at -20 °C and protect from prolonged exposure to light. To prepare a working solution, dilute 1 in 200 in 20 mM Tris buffer (pH 7.5) or buffer of choice, using 50 μL of substrate solution per assay in a 96-well plate.
