TGF-beta 1 Cell Culture Ready

A cell culture ready human recombinant protein from STEMCELL Technologies, supplied as a 0.1 mg/mL solution of dimeric, mature transforming growth factor beta 1 in 100 mM acetic acid and produced in CHO cells. The protein has a predicted molecular mass of 12.8 kDa per monomer and 25.6 kDa as the homodimer, with an accession number of P01137 (amino acids 279–390) and alternative names including cartilage-inducing factor and differentiation inhibiting factor.

Activity is determined using a SMAD/TGF luciferase reporter cell line, with an EC50 of ≤ 1 ng/mL and a representative assay value of 0.52 ng/mL; each assay is calibrated against Non WHO Reference Material (NIBSC code: 19/180). Purity is ≥ 95% by SDS-PAGE and endotoxin is measured by kinetic Limulus amebocyte lysate (LAL) analysis. The product should be stored at -20 to -80°C and is stable as supplied for at least 3 years from the date of manufacture on the label.

TGF-β1 binds to serine-threonine kinase type I and II receptors and signals through SMAD2/3 to regulate cellular functions including proliferation, differentiation, wound healing, apoptosis, and metabolism. It supports the culture of undifferentiated human embryonic stem cells and induced pluripotent stem cells alongside basic fibroblast growth factor and contributes to immune homeostasis by modulating B cell responses and mediating immunosuppressive effects on T cells and neutrophils.
