Description

Supplied by STEMCELL Technologies, this recombinant human apolipoprotein A-I (apo A-I) is a member of the apolipoprotein family and a major glycoprotein component of high-density lipoprotein (HDL) particles. Apo A-I supports reverse cholesterol transport from peripheral tissues to the liver and acts as a cofactor for lecithin cholesterol acyltransferase (LCAT) in cholesterol esterification. The protein has been shown to bind the scavenger receptors SR-B1 and SR-A1 as well as the beta chain of ATP synthase on hepatocytes, and mutations in the APOA1 gene have been associated with dysregulated HDL levels and an increased risk of amyloidosis and ischemic heart disease.

The construct corresponds to residues Asp25–Gln267 of UniProt accession P02647 and is expressed in E. coli with a polyhistidine tag and spacer sequence at the N-terminus. It has a predicted molecular mass of 30.7 kDa and a purity of ≥ 92% as assessed by SDS-PAGE under reducing and non-reducing conditions with Coomassie Blue staining. The product is lyophilized from sterile PBS with 10% glycerol (pH 7.4); trehalose (5–8%), mannitol, and 0.01% TWEEN 80 are added as protectants before lyophilization. It should be stored at -20 to -80 °C and is stable as supplied for 12 months from the date of receipt.

To prepare, centrifuge the vial before opening and reconstitute in sterile water to at least 0.25 mg/mL by pipetting the solution down the sides of the vial without vortexing. As a general guide, do not store at 2–8 °C for more than 1 month or at -80 °C for more than 3 months, and avoid repeated freeze-thaw cycles. For research use only.
