Description

From QIAGEN, this hot-start DNA polymerase is formulated as a mixture of recombinant thermostable Taq DNA polymerase from Thermus aquaticus and a highly specific monoclonal antibody that inhibits polymerization activity. The hot-start chemistry prevents polymerase activity at room temperature, improving specificity and yield for sensitive PCR assays. It enables easy setup of a hot-start PCR reaction at room temperature and is recommended for demanding applications that require highly specific amplification, including multiplex PCR and diagnostic assays.

The enzyme works rapidly and effectively across various PCR conditions, catalysing DNA synthesis in a 5′ → 3′ direction. It shows no 3′ → 5′ exonuclease activity but possesses 5′ → 3′ exonuclease activity. The antibody binds reversibly to the enzyme, suppressing polymerase activity at ambient temperatures and preventing the extension of non-specifically annealed primers and primer-dimers formed at low temperatures during PCR setup. The antibody is released from the polymerase during normal cycling conditions, so no additional incubation step is required to activate the enzyme.

The enzyme is supplied with 20 mM Tris-HCl (pH 8.0, 25°C), 100 mM KCl, 0.1 mM EDTA, 1 mM DTT, 0.5% (v/v) Nonidet P40, 0.5% (v/v) Tween 20, and 50% (v/v) glycerol. One unit is defined as the amount of enzyme required to incorporate 10 nmol of dNTPs into an insoluble DNA fraction in 30 minutes at 75°C in a 50 μL reaction. Quality control testing evaluates DNase activity and contamination by incubating 1 μg of DNA with 10 units of the enzyme for 4 hours at 37°C, with results visualised on an ethidium bromide-stained agarose gel. The product is intended for storage at frozen temperatures to maintain its hot-start performance.
