Description
A ready-to-use hot-start master mix from QIAGEN delivers high PCR specificity across a wide range of amplification applications. The premix combines HotStarTaq DNA Polymerase, a uniquely formulated QIAGEN PCR Buffer that minimizes optimization requirements, and dNTPs. By supplying all reaction components in a single mix, pipetting steps are reduced, contamination risk is lowered, and throughput and reproducibility are improved. Reactions can be conveniently set up at room temperature.
HotStarTaq DNA Polymerase is a recombinant hot-start enzyme supplied at 5 units/µl. It exhibits 5'→3' exonuclease activity, adds A overhangs, and accepts dNTP, ddNTP, dUTP, biotin-11-dUTP, DIG-11-dUTP, and fluorescent-dNTP/ddNTP substrates. The enzyme extends at 2–4 kb/min at 72 °C, with a half-life of 10 min at 97 °C and 60 min at 94 °C, and achieves amplification efficiency of ≥10^5-fold. No 3'→5' exonuclease activity, nuclease, protease, or RNase contamination, or self-priming activity is detected.
Applications include standard PCR, RT-PCR, complex genomic DNA templates, cDNA templates, very low-copy targets such as single-cell PCR, and reactions with multiple primer pairs, making the mix suitable for challenging research and high-throughput gene screening. Each batch undergoes stringent quality control, including PCR specificity and reproducibility testing, even with low-copy targets. The mix is formulated for endpoint PCR amplification of single targets from genomic DNA or cDNA.
