Taq-B DNA Polymerase (10000 U)

A thermally stable, processive 5′→3′ DNA polymerase from QIAGEN, isolated from the thermophilic organism Thermus aquaticus YT-1. The 94 kDa enzyme possesses an inherent 5′→3′ exonuclease activity and lacks a 3′→5′ proofreading function. It is well suited to routine PCR amplification of DNA fragments up to 5 kb, producing robust yields from low-complexity template sources with little or no optimisation of reaction conditions.

Quality-control specifications include a physical purity of >99% and a specific activity of 74,625 U/mg. In standard assays, single-stranded exonuclease activity is functional at 50 U, double-stranded exonuclease activity is functional at 50 U, and double-stranded endonuclease activity shows no conversion at 50 U. E. coli genomic DNA contamination is held to <10 copies per 50 U.

The enzyme is supplied in 20 mM Tris-HCl, 100 mM NaCl, 1 mM DTT, 0.1 mM EDTA, 50% glycerol and stabilizer, pH 7.5 at 25°C. It is accompanied by a 10X PCR Buffer containing 100 mM Tris-HCl, 500 mM KCl and 15 mM MgCl₂, pH 8.3 at 25°C.
