HotStarTaq Master Mix Kit (2500)

A ready-to-use master mix kit from QIAGEN designed for highly specific hot-start PCR. The HotStarTaq Master Mix contains HotStarTaq DNA Polymerase, the QIAGEN PCR Buffer, and dNTPs combined in a 2x formulation, which reduces pipetting steps and the risk of contamination while increasing throughput and reproducibility. HotStarTaq DNA Polymerase is provided in an inactive state at room temperature, preventing the formation of misprimed products and primer–dimers, and is activated by a 15-minute, 95°C incubation step that fits into standard thermal cycling programs.

The kit is supplied with the HotStarTaq Master Mix and RNase-Free Water. The master mix provides a final concentration of 1.5 mM MgCl2 in the reaction, which gives satisfactory results in most cases. A typical 50 µl reaction uses 25 µl of the 2x master mix together with primers and template DNA, and is run on a three-step cycling program (denaturation at 94°C, annealing at 50–68°C, extension at 72°C) for 25–35 cycles. HotStarTaq DNA Polymerase extends at a rate of 2–4 kb per minute at 72°C.
