Description

QIAGEN's Taq PCR Master Mix Kit is a 2x concentrated, ready-to-use solution that combines Taq DNA Polymerase, dNTPs, MgCl2 and PCR Buffer at optimized concentrations, so that only template DNA, primers and water need to be added before thermal cycling. The premixed format minimizes pipetting steps and the risk of contamination, while supporting highly reproducible PCR results. The included buffer combines KCl and (NH4)2SO4 and provides stringent primer-annealing conditions over a wider range of annealing temperatures and Mg2+ concentrations than conventional PCR buffers, reducing or eliminating the need for optimization.

The kit is designed for endpoint PCR amplification and is suitable for applications including PCR, RT-PCR and DNA fingerprinting, with single-plex reactions and genomic DNA or cDNA as starting template. The included Taq DNA Polymerase exhibits 5' to 3' exonuclease activity and adds extra A overhangs to reaction products, which is useful for cloning into U-overhang vectors. According to the manufacturer's quick-start protocol, a 100 µl reaction mix uses 50 µl of 2x master mix to deliver 2.5 units of Taq DNA Polymerase, 1x QIAGEN PCR Buffer and 200 µM of each dNTP, with the master mix providing a final MgCl2 concentration of 1.5 mM in the reaction. A simplified hot start can be performed by delaying tube placement until the cycler reaches 94°C.

Upon receipt, the kit should be stored immediately at –30 to –15°C (or –20°C per the handbook) in a constant-temperature freezer. It can alternatively be kept at 2–8°C for up to 2 months, enabling faster setup by avoiding thaw time. dNTPs are included in the master mix, and a No Template Control is recommended with every run.
