Description

The Strep-tag II is an 8-amino-acid peptide (WSHPQFEK) that binds with high selectivity to Strep-Tactin, an engineered streptavidin. QIAGEN Strep-Tactin Superflow Plus is a Superflow chromatography resin used for affinity purification of Strep-tag II-tagged proteins under physiological conditions, enabling native and active proteins to be obtained in a single step.

The Superflow Plus matrix offers a protein binding capacity of up to 9 mg/ml, representing a 3-fold increase over previous Strep-Tactin matrices. The binding affinity of the Strep-tag II to Strep-Tactin (Kd = 1 μM) is nearly 100 times higher than its affinity for streptavidin, and gentle elution of the bound protein is achieved using low concentrations of biotin or desthiobiotin, a stable, reversibly binding analog of biotin that serves as the natural ligand of streptavidin.

The resin is suitable for purifying proteins from prokaryotic and eukaryotic expression systems, including E. coli, baculovirus-infected insect cells, and transfected mammalian cells, and can be used in batch purification or with automated liquid chromatography systems. It may also be combined with a His-tag for a standardized two-step purification. Strep-Tactin matrices should be stored at 2–8°C and not frozen; under these conditions they can be stored for up to 6 months without any reduction in performance. The products are intended for molecular biology applications and are not intended for the diagnosis, prevention, or treatment of disease.
