For preparative-scale purification of transfection-grade plasmid or cosmid DNA, this gravity-flow kit from QIAGEN uses anion-exchange technology to process culture volumes of 2.5–5 liters, with yields of up to 10 mg of plasmid DNA per run.

The kit is processed manually by gravity flow at a throughput of one sample per run, with an estimated preparation time of 320 min. It supports high-copy, low-copy, and cosmid plasmid types, and purified DNA is suitable for transfection, cloning, sequencing, and capillary sequencing applications. Prepacked QIAGEN-tip anion-exchange columns operate by gravity flow, avoiding the use of toxic substances such as phenol, chloroform, ethidium bromide, and CsCl.

Lysate clearing is performed by centrifugation, and the eluted DNA is concentrated and desalted by isopropanol precipitation. As an optional step, the kit can be combined with QIAfilter Mega-Giga Cartridges for rapid clearing of bacterial lysates by filtration instead of centrifugation.
