QIAGEN Plasmid Mega Kit (5)

Designed for purifying transfection-grade plasmid or cosmid DNA, this gravity-flow kit from QIAGEN uses anion-exchange tips as part of the manufacturer's family of plasmid DNA extraction kits. The kit uses manual processing with gravity flow through a QIAGEN-tip, selectively binding plasmid DNA under appropriate low-salt and pH conditions while RNA, proteins, and other low-molecular-weight impurities are removed by a medium-salt wash. Pure plasmid DNA is then eluted in high-salt buffer, concentrated by isopropanol precipitation, and collected by centrifugation.

Suitable for high-copy, low-copy, and cosmid DNA, the kit accommodates culture volumes ranging from 500 ml to 2.5 liters depending on plasmid copy number, and yields up to 2.5 mg of plasmid DNA per preparation, with a typical run time of 220 min. One sample is processed per run. Purified DNA is highly suitable for applications including transfection, cloning, sequencing, and capillary sequencing, and the system avoids toxic substances such as phenol, chloroform, ethidium bromide, and CsCl. As an optional step, QIAfilter Mega-Giga Cartridges can be used for rapid clearing of bacterial lysates by filtration instead of centrifugation, operating with house vacuum.
