QIAGEN Plasmid Mega Kit (25)

The QIAGEN Plasmid Mega Kit is a gravity-flow, anion-exchange system designed for purification of transfection-grade plasmid DNA. It uses prepacked QIAGEN anion-exchange tips that operate by gravity and never run dry, delivering DNA purity equivalent or superior to that obtained by two successive rounds of CsCl gradient centrifugation, while avoiding toxic substances such as phenol, chloroform, ethidium bromide, and CsCl.

Recommended maximum culture volumes are 500 ml for high-copy plasmid and 2.5 liters for low-copy plasmid, with yields of up to 2.5 mg of high-copy plasmid DNA per prep. The kit is processed manually, one sample per run, using anion-exchange technology, with a typical time per run of approximately 220 minutes. Kits can be stored at room temperature (15–25°C) for at least 2 years if not otherwise stated on the label.

Plasmid DNA prepared with the kit is suitable for high-copy, low-copy, and cosmid DNA types, and supports applications including transfection, cloning, sequencing, capillary sequencing, and in vitro transcription. As an optional protocol step, the kit can be combined with QIAfilter Mega-Giga Cartridges for rapid clearing of bacterial lysates by filtration instead of centrifugation.
