Smoothened Agonist (SAG) is a potent small-molecule activator of the Smoothened (Smo) seven-transmembrane receptor, the central signal transducer of the Hedgehog (Hh) signaling pathway. In stem cell biology, SAG is widely used to drive ventral forebrain and midbrain patterning, supporting protocols that generate floor-plate identities, dopaminergic neurons, and patterned neural organoids from human pluripotent stem cells.

Each vial from STEMCELL Technologies contains 10 mg of lyophilized SAG, supplied for routine Hedgehog pathway activation studies. The compound is reconstituted in DMSO to a working stock and typically diluted into culture medium at nanomolar to low micromolar concentrations, depending on the desired differentiation outcome.

For stability, the lyophilized material should be stored at -20 °C, protected from light, and the reconstituted stock aliquoted to minimize freeze-thaw cycles. Under these conditions the reagent retains activity for downstream cell culture and in vitro applications.

Typical uses include neural induction and patterning of human ES and iPS cells, dopaminergic neuron differentiation, organoid and tissue-engineering workflows, and mechanistic studies of Smoothened-mediated signaling in development and disease research. This 10 mg format suits laboratories running multiple differentiation experiments or long-term Hh pathway studies, and the product is supplied for research use only.
