STEMCELL Technologies supplies a rat monoclonal IgM, kappa isotype control antibody (Clone RTK2118) designed to estimate the degree of non-specific binding by an antigen-specific antibody. The unconjugated format is offered, and conjugated variants including Alexa Fluor® 488, APC, Biotin, FITC, and PE are also available from the same clone. According to the manufacturer, the isotype control should ideally share the same heavy chain subclass and light chain as the specific antibody being used, and where a conjugated primary antibody is employed, an isotype control conjugated to the same fluorochrome should be selected.

The use of an appropriate isotype control helps confirm the specificity of the antigen-specific antibody and reveals non-specific binding that may result from interactions with Fc receptors or other cell components. Clone RTK2118 has been verified for assessing non-specific binding in flow cytometry and immunofluorescence microscopy applications, including both surface and intracellular staining.
