Rosiglitazone, 50 mg

A potent and selective ligand for peroxisome proliferator-activated receptor gamma (PPARγ) supplied by STEMCELL Technologies as a crystalline solid. The compound binds to the PPARγ ligand-binding domain with a Kd of 43 nM and activates luciferase-based expression constructs PPARγ1 and PPARγ2 with EC₅₀ values of approximately 30 nM and 100 nM, respectively. It has a molecular weight of 357.4 g/mol, a purity of ≥ 98%, and is also known as Avandia or BRL 49653 (CAS 122320-73-4). Demonstrated solubility reaches ≤ 2.7 mM in absolute ethanol and ≤ 95 mM in DMSO. The product is stable at -20°C as supplied when protected from prolonged exposure to light, and remains stable for 12 months from the date of receipt. Stock solutions should be prepared fresh before use; for cell culture applications, stock should be diluted into culture medium immediately before use, avoiding final DMSO concentrations above 0.1% due to potential cell toxicity. The compound is also used as a medium for the differentiation of human MSCs into adipocytes and as a supplement (50X) for the serum-free culture of neurons.

Published research applications include stimulating embryonic mouse neural stem cell proliferation while inhibiting neuronal differentiation, inducing adipocyte differentiation in C3H10T1/2 stem cells, promoting endothelial differentiation and inhibiting smooth muscle differentiation in angiogenic progenitor cells, stimulating adipocyte differentiation and decreasing osteogenesis in mouse and human bone marrow-derived mesenchymal stem cells, and enhancing osteoclast formation in mouse bone marrow cells stimulated with RANKL and M-CSF.
