Description
Manufactured by STEMCELL Technologies, the RosetteSep™ Human Total Lymphocyte Enrichment Cocktail is an immunodensity negative selection reagent designed to enrich lymphocytes directly from whole blood. Unwanted cells are crosslinked to red blood cells via Tetrameric Antibody Complexes recognizing CD16, CD36, CD66b and glycophorin A, allowing purified lymphocytes to be collected at the interface between plasma and density gradient medium following standard centrifugation.
The protocol involves a 20 minute incubation of the sample with the cocktail, followed by layering on a buoyant density medium such as RosetteSep™ DM-L, Lymphoprep™, or Ficoll-Paque™ and centrifugation for 20 minutes. The enriched fraction typically contains 94 ± 2% CD2+ and CD19+ cells when starting with fresh peripheral blood. The cocktail includes mouse monoclonal IgG1 antibodies against human CD2 and CD19.
Designed for use with human whole blood or buffy coat, the workflow requires no special equipment or training and yields untouched, viable cells. It can be combined with SepMate™ tubes for consistent, high-throughput sample processing. A published case study from the University of Maryland School of Medicine reported that the RosetteSep™ and SepMate™ system reduced NK cell isolation time from four hours to one hour for a 450 mL unit of blood.
