For immunology researchers studying type 2 inflammation, allergy, and tissue-resident immunity, this sample-size vial from STEMCELL Technologies isolates innate lymphoid cells type 2 (ILC2) directly from human whole blood or peripheral blood mononuclear cells (PBMCs) using immunodensity negative selection. A human-specific antibody cocktail crosslinks unwanted leukocytes—including T cells, B cells, NK cells, monocytes, and granulocytes—to red blood cells through tetrameric antibody complexes, forming dense rosettes that pellet during density gradient centrifugation. The untouched ILC2 fraction is recovered at the interface, ready for downstream flow cytometry, single-cell sequencing, or functional studies without the need for a magnetic column.

The vial supports approximately 1–2 mL of fresh whole blood or up to 1 × 10⁷ mononuclear cells per separation, with a total processing time of roughly 25 minutes when combined with Lymphoprep. Purity and recovery have been validated across healthy donor samples, and the protocol integrates cleanly with standard workflows including intracellular cytokine staining and transcription factor profiling.

Each vial is shipped refrigerated and should be stored cold until first use to preserve antibody activity through the expiration date on the label. Researchers commonly pair the enrichment step with downstream assays such as flow cytometry, RNA sequencing, and in vitro culture of ILC2 to study mucosal immunity, helminth infection, and allergic airway disease.
