RosetteSep HLA B Enrich Cocktail, 10mL

A cell enrichment cocktail from STEMCELL Technologies is designed for the in vitro isolation of untouched B cells from human whole blood by negative selection. Unwanted cells are crosslinked to red blood cells (RBCs) via a combination of monoclonal antibodies in phosphate-buffered saline (PBS), forming immunorosettes that pellet with free RBCs when centrifuged over a density gradient medium. Desired cells are never labeled with antibody and are easily collected as a highly enriched population at the interface between the plasma and the density gradient medium, making them suitable for flow cytometry crossmatch assays or serology.

The cocktail is for in vitro diagnostic use and should be stored at 2 - 8°C (it may be shipped at 15 - 25°C but should be refrigerated on receipt and not frozen). In use, the cocktail is added at 40 µL/mL of whole blood, the sample is incubated for 20 minutes at room temperature (15 - 25°C), and then centrifuged at 1200 x g for 20 minutes at room temperature with the brake off over a density gradient medium with a density of 1.077 g/mL. A precipitate may be observed in the vial but will not affect performance.

According to manufacturer data, the typical B cell (CD19+) purity following enrichment is greater than 65%, with the CD19+ content of the enriched fraction typically ranging from 59.3% to 97.2% when starting from whole blood. Purity can be assessed by flow cytometry using an Anti-Human CD19 antibody, Clone HIB19, or an Anti-Human CD20 antibody, Clone 2H7. The product is manufactured under a quality management system certified to ISO 13485 and is intended for professional users observing good laboratory practice; once added to human cells the suspension should be treated as potentially biohazardous, and the cocktail is a potential irritant to eyes, respiratory system, and skin.
