RosetteSep Granulocyte Depletion, 2mL

Designed to deplete granulocytes from whole human blood, the RosetteSep™ Human Granulocyte Depletion Cocktail from STEMCELL Technologies uses tetrameric antibody complexes (TAC) that bind unwanted granulocytes via recognition of CD66b, as well as red blood cells via glycophorin A. When centrifuged over a density gradient medium such as Lymphoprep™ or Ficoll™, the labelled granulocytes form a pellet together with the red blood cells, while the granulocyte-depleted fraction is recovered as an enriched population at the plasma and density medium interface.

The cocktail supports the immunomagnetic isolation of untouched neutrophils directly from whole blood. According to the manufacturer's protocol, performing immunodensity cell separation with this product before density gradient centrifugation can yield a sample containing < 1% granulocytes, compared to 20% granulocytes with density gradient centrifugation alone. The approach is particularly useful for older human whole peripheral blood samples (> 24 hours post collection), where changes in granulocyte density can otherwise cause granulocyte contamination of the mononuclear cell fraction when using a density gradient medium alone.

A SepMate™ tube may be used in combination to harvest isolated mononuclear cells with a simple pour. Before beginning the procedure, the whole blood sample, the cocktail, PBS + 2% FBS, Lymphoprep™, and centrifuge should all be at room temperature (15 - 25ºC).
