Description

Developed by STEMCELL Technologies, this depletion cocktail enriches epithelial circulating tumor cells (CTCs) from human whole blood by depleting CD45+ cells. The antibody cocktail crosslinks unwanted hematopoietic cells to multiple red blood cells (RBCs) in the sample, forming immunorosettes. This increases the density of the unwanted (rosetted) cells so that they pellet along with the free RBCs when centrifuged over a density gradient medium, while the desired cells are easily collected as an enriched population at the interface between the plasma and the density gradient medium.

Isolated cells are untouched and viable, and are immediately ready for downstream applications such as flow cytometry, cell culture, or DNA/RNA extraction. The cocktail contains a combination of monoclonal antibodies in PBS, including a mouse monoclonal IgG1 antibody against human and chimpanzee CD45, and should be stored at 2 - 8°C without freezing. It can be combined with SepMate™ tubes for faster, more consistent high-throughput sample processing. The cocktail is fast and easy-to-use, and requires no special equipment or training.

For optimal performance, whole peripheral blood collected within the last 24 hours and stored at room temperature (15 - 25°C) is recommended. The concentration of nucleated cells in the sample should not exceed 5 x 10^7 cells/mL, with a minimum ratio of 100 RBCs per nucleated cell when working with sources such as buffy coat. The recommended medium is Dulbecco's PBS containing 2% fetal bovine serum, and the recommended density gradient medium is Lymphoprep™ or another medium with a density of 1.077 g/mL.
